Top-up doses were calculated as half the initial dose of medetomidine, ketamine and butorphanol. During surgery, mean SpO2 was 78 9 ; %, and mean pulse rate was 54 4 ; beats minute. All animals in group # 1 had episodes when SpO2 dropped below 80%. In group # 2, the mean induction time was 7.8 2.6 ; minutes, and the mean surgery time was 15.6 6.1 ; minutes. Induction was calm, muscle relaxation was good throughout the procedure and the beavers did not seem to react to sound stimuli. Mean SpO2 was 84 4 ; % and mean pulse rate during surgery was 54 7 ; beats minute. Brief episodes when SpO2 dropped below 80% occurred in seven animals, which were then allowed to breathe 100% oxygen through a face mask held over the animals' noses. All surgeries were successfully performed, and no specific problems were encountered neither during surgery, in the postoperative period nor before the beavers were released back into the water at the site of capture.

Abstract: Yaji, otherwise `suya sauce', used in serving the meat delicacy called `Suya', is a complex mixture of spices and additives whose active ingredients on individual basis, are known to have side effects if consumed in excess. The growing concern is that the excessive consumption of Yaji signifies an excessive consumption of a combination of these constituents amongst which are ginger, cloves, red pepper and black pepper; which are mixed without a standardized format. This study was therefore designed to correlate the individual with the combined effects of the excessive consumption of these spices on the histology of the liver of adult rabbits. Involved in this study were 12 rabbits that were divided into six groups of two 2 ; each A, B, C, D, E and F ; . Those in groups B, C, D, E and F, constituted the test groups whereas group A served as the control. For 21 days, test group B were fed with a combination of Ginger 3g ; , Clove 3g ; , red Pepper 3g ; and Black Pepper 3g ; per day while test groups C, D, E and F were fed with ginger 3g ; , Clove 3g ; , Red pepper 3g ; and Black Pepper 3g ; per day respectively. The histological observations show that the excessive consumption of these spices can cause necrosis of liver hepatocytes and therefore acute hepatitis. Key words: Yaji, spices, liver, hepatocytes, necrosis and cognex.

With the steady increase in life expectancy cancer continues to grow into a major health problem. An estimated 35% of human cancer-related mortality is attributed to diet 1 ; . While the diet is a source of carcinogens it is also the source of inhibitors of the carcinogenic process. Laboratory based research over the past few decades has established an important role for fruits and vegetables as cancer risk reducers either by blocking cancer initiation or suppressing promotion 2 ; . More recently several groups have shown that phytochemicals present in spices also specifically target pathways that are deregulated in cancer. Since multiple pathways are deregulated in cancer the consumption of a variety of plant products improves the chances of targeting multiple deregulated pathways in cancer. For example compounds such as eugenol from cloves and bay leaves ; , curcumin turmeric ; , capsaicin peppers ; , gingerol ginger ; , and diallyl sulfide garlic ; have been found to regulate specific pathways involved in cell cycle progression, apoptosis, survival, intracellular signaling, as well as cellular detoxification 3 ; . Melanoma Melanoma is the deadliest form of skin cancer accounting for almost 78% of skin cancer fatality. It is one of the fastest growing cancers in the developing world. The 5-year recurrence rates of patients with high-risk melanoma are 5580% and survival rates are poor 4 ; . Adjuvant therapy using interferon given to stages II, III, and IV following surgery produce a response rate of 1020% but only for a short duration 3 9 ; . Chemotherapeutic agents such as dacarbazine DTIC ; show a response rate of 1520% and a combination of cisplatin and dacarbazine with tamoxifen and carmustine produce a response rate of 55% 4 10 ; . However, the typical short life of the response rates has had no significant effect on overall survival. Ongoing and completed clinical trials using monoclonal antibodies and vaccine therapy for malignant melanoma treatment have been promising but overall have not made an impact on malignant melanoma mortality rates. Hormonal therapy of disseminated melanoma has been used infrequently and is generally regarded as being ineffective, although use of NSC-17256 a drug with glucocorticoid and anti-estrogenic activities ; produced objective tumor regression in 5 of patients 5 11 ; . Dietary Agents for Melanoma Chemoprevention Thus far, not many dietary agents have been tested for chemopreventive activities against malignant melanoma although there are many potential agents that target specific pathways deregulated during the development of malignant melanoma. In almost all these cases provocative preclinical data has been provided but no randomized trials have been undertaken to carefully evaluate the pathways targeted, establish dose-response relationships, validation of biomarkers, etc. Use of Eugenol as a Botanical Chemopreventive Agent 4-allyl-2-methoxyphenol, also known as Eugenol, is found in reasonable quantities in the essential oils of different spices such as Syzgium aromaticum clove ; , Pimenta racemosa bay leaves ; , and Cinnamomum verum cinnamon leaf ; . These spices are used as flavoring agents in culinary practices all over the world. Further, eugenol has been used as an antiseptic, anti-bacterial, analgesic agent in traditional medical practices in Asia. We have shown that eugenol exhibits potent anti-proliferation activity in melanoma cells that represent different stages of progression. We have also shown that eugenol blocks cells in the S phase of progression; induces apoptosis; and inhibits E2F1 transcriptional activity in melanoma cells. Further overexpression of E2F1 was able to overcome the growth inhibition mediated by eugenol 5 ; . Eugenol also inhibited the invasion of melanoma cells through the basement membrane in Boyden chamber assays. In the B16 xenograft model intraperitoneal delivery of eugenol inhibited melanoma metastasis to the lungs compared to untreated controls. These results suggest that it is possible to use dietary agents and botanicals for the inhibition of malignant melanoma. In the case of melanoma, a better approach may be to screen such phytochemicals in a secondary prevention setting to prevent the progression of easily treatable primary disease to the highly aggressive metastatic disease that is extremely resistant to current therapeutic regimens.

Moreover, evidence that older aged patients tend to tolerate chemotherapy better than younger patients was first attributed to the less aggressive treatment regimens administered to the elderly. Today, however, older patients frequently receive aggressive therapy.48, 49 A comparison of two groups of patients younger than 65 and older than 65 years of age ; with breast cancer who were receiving similar chemotherapy protocols found that with only one exception, younger patients consistently reported greater problems with NVR.31 Since women 50 years of age or younger are more likely to experience NVR, they are also more prone to anticipatory NVR.50 A recent review found that up to 90% of pregnant women experience NVR.51 Women who were queried prior to their initial chemotherapy treatment about their past experiences of NVR e.g., with stomach flu, motion sickness, surgical procedures, pregnancies ; and reported high levels subsequently described more episodes of NVR than did those with less remarkable NVR histories.27.

E2 levels, 10 000 IU of hCG Profasi; Serono ; was given. Oocytes were retrieved 34 h later under the guidance of vaginal ultrasound or laparoscopy. The oocyte cumulus complexes were cultured in human tubal fluid HTF ; medium with 15% heat-inactivated maternal serum and incubated at 378C in an atmosphere of 5% CO2 in air. Cryopreservation of part or all of the oocytes For infertile couples, six to 12 oocytes were generally used for IVF or ICSI. The additional mature oocytes were cryopreserved. For couples having unexpected ejaculatory failure or unmarried women, all of the mature oocytes were frozen. The cumulus cells were dissected using 23 G needles followed by brief exposure 30 s ; to hyaluronidase 80 IU ml; Sigma; USA ; . The corona cells were completely removed by pipetting through micropipettes. The oocytes were checked for the presence of the first polar body in the perivitelline space metaphase II ; . Immature metaphase I or germinal vesicle ; stage oocytes were not frozen. Freezing was routinely initiated within 2 3 h post-retrieval. Preparation of pretreatment, freezing, and dilution solutions The solutions for cryopreservation and dilution were prepared using Dulbecco's phosphate-buffered saline DPBS ; Gibco, USA ; plus 20% maternal serum. The pretreatment solution consisted of 1.5 mol l PROH Sigma ; . The freezing solution contained 1.5 mol l PROH plus 0.3 mol l sucrose. For thawing procedures, the oocytes were thawed in stepwise dilution solutions with 1.0 mol l PROH plus 0.3 mol l sucrose, 0.5 mol l PROH plus 0.3 mol l sucrose, and 0.3 mol l sucrose. Equilibration and slow freezing of oocytes Fabbri et al., 2001 ; The oocytes were transferred into 0.8 ml of pretreatment solution and kept at room temperature 22 248C ; for 10 min. Then the oocytes were transferred to the freezing medium. After the 15 min of equilibration, the oocytes were loaded into a 0.25 ml plastic straw IMV, France ; and transferred into an automated Kryo 10 series III biological vertical freezer Planer Products Ltd, UK ; . The initial chamber temperature was 208C, then slowly reduced to 278C at a rate of 28C min. Ice nucleation was induced manually at 2 78C. After a holding time of 10 min at 278C, the straws were cooled slowly to 2308C at a rate of 0.38C min and then rapidly to 2 1508C at a rate of 508C min. After 10 12 min of temperature stabilization, the straws were transferred into liquid nitrogen tanks and stored until thawing. Treatment cycles with frozen thawed oocytes For thawing cycles, endometrial preparation involved the natural ovulatory cycle or the hormonal replacement cycle. For the protocol of hormonal replacement cycle, the female received oral estradiol valerate 2 mg ; with incremental dosages of 4 mg from day 3 to day 8; mg from day 9 to day 11; and then 12 mg from day 12. On day 14, serum E2, LH and progesterone were tested and endometrial thickness was measured using vaginal ultrasonography. If the endometrial thickness was mm, with no evidence of ovulation, progesterone in oil 50 mg daily ; was administered i.m. The cryopreserved oocytes were thawed and ICSI was performed at the day of starting progesterone administration. If pregnancy was achieved, the hormonal replacement was carried out to 12 weeks of gestation. To thaw, the straws were air-warmed for 30 s and then immersed in a 308C water bath for 40 s until all traces of ice disappeared Fabbri et al., 2001 ; . The contents of the straws were expelled in 1.0 mol l PROH plus 0.3 mol l sucrose solution and the oocytes and colestipol.
