Electron microscopic imaging of unstained biological macromolecules in the frozen hydrated state have been utilized. Translational and rotational image alignment, averaging and Fourier filtering of two independent subsets of the images showed the surface of the particles to have a sub-structure which may reflect the organization of apoprotein B and phospholipid at the surface. In separate studies monoclonal antibodies to apoB bound to LDL could be visualized at the particle surface. More extensive averaging and processing techniques currently being applied to these images will provide a more detailed description of the structure of LDL and the organization of apoB at the particle surface.
We have used a new family of zinc-specific-responsive fluorescent dyes ZPs ; to study the sequestration and secretion of zinc from Paneth cells, which are located in the bases of the crypts of Lieberkuhn within the rat small intestine. Vivid ZP fluorescence zinc staining of Paneth cell secretory granules is seen in both cryostat sections and isolated crypts, providing firm evidence for a pool of labile rapidly exchangeable ; zinc within these cells. We further demonstrate that this ionic zinc pool is secreted under physiological conditions. In vivo stimulation of the small intestine by IP injection of the secretagogue pilocarpine results in discrete zinc staining within the lumens of subsequently isolated crypts, concomitant with a decrease in the zinc staining of Paneth cell granules located within the same crypts. In contrast, the secretion of zinc into the lumens of isolated crypts stimulated in vitro with either carbachol or LPS lipopolysaccharide ; is not observed. However, a distinct change in Paneth cell morphology, suggesting attempted secretion, is seen in response to the direct application of cholinergics but not LPS. These findings suggest that zinc is coreleased with other Paneth cell anti-microbials, and that the intact intestine is necessary for secretion into the crypt lumen. Administration--In the administration of this agent the characteristic results may be obtained almost equally well, either from an infusion of the leaves, or from any of the preparations named. The hypodermic administration of the alkaloid pilocarpine, is preferable in many cases, especially where the promptness of its action is desired. The liquid preparations are often unacceptable to a disordered or sensitive stomach and then minute doses of the alkaloid in pellets or granules will be, found a most desirable form for administration. The action of the fluid extract or tincture of jaborandi and impure pilocarpine is sometimes disappointing, failing entirely to produce their characteristic influence and perhaps producing results contrary!
